ABSTRACT

Objective:To explore the prognostic value of combined fibrinogen/albumin ratio (FAR) and CA724 in patients with stage Ⅱ/Ⅲ gastric cancer after radical resection. Methods:A total of 932 patients were enrolled in the study, and the best cut-off values of CEA, FAR, NLR and other variables were obtained through ROC curve analysis. According to the FAR-CA724 score, patients were divided into 3 groups: FAR-CA724=0 (CA724<3.43 ng/ml and FAR<0.083), FAR-CA724=1 (CA724≥3.43 ng/ml and FAR≥0.083) and FAR-CA724=2 (CEA≥3.43 ng/ml and FAR≥0.083).Results:After FAR-CA724 grouping, the patient's age (χ 2=12.02, P=0.002), gender (χ 2=15.91, P<0.001), tumor size (χ 2=18.22, P<0.001), hypertension (χ 2=6.35, P=0.042), tumor location (χ 2=26.09, P<0.001), degree of differentiation (χ 2=12.46, P=0.002) and pTNM staging (χ 2=6.68, P=0.035) are significantly different. Survival analysis showed that there were significant differences in OS between the three groups of patients (FAR-CA724=0, 1, and 2: 88.2%, 64.3% and 37.8%, respectively, P<0.001). By multivariate analysis FAR-CA724 is an independent risk factor affecting OS in patients with stage Ⅱ/Ⅲ gastric cancer after radical surgery. Conclusions:Preoperative FAR-CA724 may be a potential blood marker for predicting the prognosis of GC patients.

ABSTRACT

BACKGROUND:Buccal Multiloop Removable Appliance can interceptively correct mutiple adolescent malocclusions. But the clinical problem of Buccal Multiloop fatigue fracture is not solved yet. How to prolong the fatigue fracture cycle is stil in the research stage. OBJECTIVE: To study the effect of different temperature of heat treatment on the Buccal Multiloop fatigue fracture cycle, thereby to select a relatively optimal method to enhance the fatigue fracture cycle. METHODS: Thirty-five left HL-2 Buccal Mltiloops were divided into seven groups according to different ways of heat treatment. Each group consisted of five samples. They were an untreated group, three pre-bending groups (320, 420, 520℃ heat treatment before bending) and three post-bending groups (320, 420, 520℃ heat treatment after bending). The dental stainless steel wires and Buccal Multiloop were respectively treated by low-temperature annealing. The data were recorded and evaluated after the samples tested by the 3D Simulating Movement of TMJ Testing Machine. The features of fatigue fracture were observed by scanning electron microscope. RESULTS AND CONCLUSION: The mean values of the Buccal Multiloop fatigue fracture cycle from largest to smalest were as folows: 520℃ pre-bending group > 420℃ pre-bending group > 320℃ pre-bending group > untreated group > 520℃ post-bending group > 320℃ post-bending group > 420℃ post-bending group. The fatigue fracture cycle of Buccal Multiloop made of the dental stainless steel wires after 520℃ annealing treatment was longer than others. By the observation of scanning electron microscope, the fracture crack extension area had the tendency to expand, transient interruption was delayed and the tissue structure became more uniform.

ABSTRACT

BACKGROUND:The self-ligating bracket is superior to the conventional bracket in orthodontic treatment, but there are a lot of controversies. At present, researchers have different views on the function of self-ligating brackets. OBJECTIVE:To summarize the comparative study of self-ligating brackets and conventional brackest with the direction and development in recent years. METHODS:A computer-based search of PubMed (2000-2014) and CNKI (2000-2014) databases was done for relevant articles, using the key words of “self-ligating brackets; conventional orthodontic brackets” in English and Chinese, respectively. RESULTS AND CONCLUSION:Self-locking brackets saves chair side time, which has been confirmed and accepted. There are different views for the issue that self-ligating brackets can reduce friction in orthodontic treatment. It is widely accepted that, the self-ligating brackets using thin round wire had less friction than the conventional brackets; whether the friction of self-ligating brackets in the closed gap and fineness adjustment stages is less than the conventional brackets stil remains unclear, which need further clinical comparative studies. The self-ligating brackets showed no significant difference with the conventional brackets in arch expansion, reducing anchorage loss and root adsorption. The majority of relevant studies is performedin vitro, and cannot exactly simulate the dental conditions. A large-size experiment with the uniform criteria for therapeutic efficacy is needed to objectively evaluate self-ligating brackets.

ABSTRACT

Objective To observe the effect of silencing P2X7 receptor (P2X7R) by RNA interference on microglial releasing interleukin 1β (IL-1β),tumor necrosis factor-α (TNF-α) and nitric oxide (NO).Methods The small interfering RNA (siRNA) targeting P2X7R gene was observed. The primary microglial cells activated by amyloid-β (Aβ-42) were infected with the lipofectaminesiRNA.The groups were designated as Aβ-1-42,Aβ1-42/siNC,Aβ1-42/siP2X7R and blank control groups according to the different stimulus. After RNA interference for 48 hours, the microglial cells were collected. The survival rate of microglia was detected by CCK-8.The levels of P2X7R mRNA and protein were detected by Real-time PCR and Western blotting respectively. The microglial morphology and the P2X7R immune response were observed by immunocytochemistry staining. Then levels of IL-1β,TNF-α and NO in the supernatantwere measured. Results The Aβ1-42 and siRNA had no effects on the survival rate of microglia. After RNA interference silencing P2X7R,the expression levels of P2X7R mRNA and protein in Aβ1-42/siP2X7R group were decreased significantly, and in this group, the microglial activity and P2X7R immune response were all reduced. In Aβ1-42,Aβ1-42/siNC,Aβ1-42 /siP2X7R and blank control groups, the supernatant levels of IL-1β were (52.54±3.21) μg L,(54.94±2.54) μg/L,(28.70±3.58) μg/L,(24.55±4.34) μg/L,respectively and the supernatant levels of TNF-α were (64.40±4.80) μg/L,(60.94±1.63) μg/L,(25.69±3.13) μg/l,(19.21±1.97)μg/L,respectively.As compared with Aβ1-42 and Aβ-42/siNC groups, the levels of IL 1β and TNF-α in Aβ1-42 /siP2X7R group was decreased significantly (P< 0.05),and IL-1β level had no significant difference between Aβ1-42/siP2X7R and blank control groups. The same result was observed in the level of NOin the supernatant. The levels of NO were (1.33±0.19) μmol/L,(4.49±0.28) μmol/L,(4.78±0.33) μmol/L and (1.07±0.36) μmol/L in Aβ1-42/siP2X7R,Aβ1-42,Aβ1-42 /siNC and blank control groups respectively. Conclusions The silencing expression of P2X7 R by RNA interference effectively decreases the levels of IL-1β,TNF-α and NO released by microglia.P2X7 R can be used as an effective therapeutic target for RNA interference treatment of Alzheimer's disease.

ABSTRACT

OBJECTIVE: The prior functional MRI studies have demonstrated significantly abnormal activity in the bilateral superior temporal gyrus (STG) of anxiety patients. The purpose of the current investigation was to determine whether the abnormal activity in these regions was related to a loss of functional connectivity between these regions. MATERIALS AND METHODS: Ten healthy controls and 10 anxiety patients underwent noninvasive fMRI while actively listening to emotionally neutral words alternated by silence (Task 1) or threat-related words (Task 2). The participants were instructed to silently make a judgment of each word's valence (i.e., unpleasant, pleasant, or neutral). A coherence analysis was applied to the functional MRI data to examine the functional connectivity between the left and the right STG, which was selected as the primary region of interest on the basis of our prior results. RESULTS: The data demonstrated that the anxiety patients exhibited significantly increased activation in the bilateral STG than the normal controls. The functional connectivity analysis indicated that the patient group showed significantly decreased degree of connectivity between the bilateral STG during processing Task 2 compared to Task 1 (t = 2.588, p = 0.029). In addition, a significantly decreased connectivity was also observed in the patient group compared to the control group during processing Task 2 (t = 2.810, p = 0.012). CONCLUSION: Anxiety patients may exhibit increased activity of the STG but decreased functional connectivity between the left and right STG, which may reflect the underlying neural abnormality of anxiety disorder, and this will provide new insights into this disease.

ABSTRACT

Objective To label neural stem cells (NSCs) with superparamagnetic iron oxides (SPIO) and to explore the tropism of NSCs after transplantation into the hippocampus of APP/PS1 AD mice by MRI.Methods NSCs from C57BL/6 mouse were cultured and identified.Feridex and Poly-L-Lysine were added into the medium to be co-cultured to make magnetic labeled NSCs and transmission electron microscopy was used to identify the iron particles in NSCs.Transgenic (tg) and wild-type (wt) mice at 12 months of age were divided into three groups: SPIOs labeled NSCs group (A and C),unlabeled NSCs group(B).Feridex-labeled NSCs were migrated into the hippocampus of APP/PS1 AD mice to monitor in vivo by MRI.After 1,2,4 and 6 weeks, the mice were sacrificed and their brain tissues were sectioned to investigate the migration of SPIO labeled NSCs and compared with MRI.Results NSCs of C57BL/6 mice were cultured successfully.Transmission electron microscope showed visible iron granules in cytoplasm.MRI detection of labeled cells: T2WI and T2* WI showed remarkable low signal intensity at the hippocampus injection points 1 week after transplantation, particularly on T2* WI.Area of low signal intensity enlarged increasingly along the injection points after 2 weeks. At 4 weeks, area of low signal intensity spread throughout the hippocampus, but intensity shadowed Six weeks later, low signal intensity almost disappeared. There was no obvious low signal change in unlabeled cell transplantation group. For wt mice, size and location of low signal did not appear obvious change at all designated time points.Prussian blue positive cells were observed in the hippocampus, indicating that NSCs labeled with SPIO could survive, migrate and differentiate in the brain of the APP/PS1 AD mice.Changes of pathology were well correlated with the area where a signal intensity loss was observed in MRI 1,2,4 and 6 weeks after transplantation Conclusions Diffuse migration of transplanted NSCs labeled with SPIO is observed in the hippocampus in APP/PS1 tg mice, and MRI technique is an ideal method for tracking labeled stem cells after grafting in vivo.

ABSTRACT

Objective:This study aims to investigate the expression pattern of miR-512-3p in breast cancer and noncancerous paired specimens as well as the effects of miR-512-3p on the proliferation, apoptosis, cell cycle, and cloning of MD-MBA-231 breast cancer cells. The study also aims to identify the miR-512-3p target gene. Methods:Reverse transcription-polymerase chain reaction (RT-PCR) was conducted to quantify the miR-512-3p expression in breast cancer and noncancerous paired specimens. Methylthiazol tetrazolium (MTT) assay, flow cytometry, and clone formation assay were used to characterize the function of miR-512-3p in breast cancer. Target prediction was performed using the TargetScan, PicTar, and miRanda software. The results were validated using RT-PCR and western blot target validation. Results:The relative expression of miR-512-3p in breast cancer specimens was significantly lower than those in normal breast specimens (P<0.05). MTT assay revealed that 48 h after transfection, miR-512-3p significantly repressed the proliferation of MD-MBA-231 cells at a suppression rate of 45.38%and at a concentration of 100 nmol/L. MiR-512-3p increased the percentage of early apoptotic cells in the treatment groups (9.32 ± 0.41)%compared with those in the blank controls (3.1 ± 0.54)%and in the negative controls (2.9 ± 0.39)%(P<0.05). Significant differences were found in the percentages of the G0/G1-and G2/M-phase cells after miR-512-3p transfection compared with those in the controls (P<0.05). In the cloning assay, clone formation was inhibited in the miR-512-3p-transfected groups compared with those in the control groups. RT-PCR and western blot results indicate that miR-512-3p significantly inhibited the c-FLIP mRNA and protein expression. Conclusion:MiR-512-3p expression is relatively decreased in breast cancer specimens compared with those in the normal samples. The negative effect of miR-512-3p on cell proliferation and clone formation and its positive effect on early apoptosis through c-FLIP targeting suggest that miR-512-3p acts as a tumor suppressor gene in breast cancer. Therefore, miR-512-3p may be a new target for the diagnosis and treatment of breast cancer in the future.

ABSTRACT

BACKGROUND:In the treatment of teeth later fixed appliance, whether the expansion effect can be maintained? Whether the expansion in the process of mandibular rotation could be improved? Whether the high angle cases are suitable for rapid palatal expansion? OBJECTIVE:To sum the effect of rapid maxil ary expansion combined with fixed orthodontic technique on the occlusion craniofacial three-dimensional direction, hard and soft tissues and temporomandibular joint, as wel as its long-term stability. METHODS:The CNKI database, PubMed database and Wanfang database were searched for the academic journals and literatures on rapid maxil ary expansion combined with fixed appliance with the key words of“maxil ary expansion, fixed appliance, high angle, stable”. The analysis function of database was used. RESULTS AND CONCLUSION:Rapid maxil ary expansion can effectively obtain arch expansion, increase the arch width, and provide space of align teeth required. Fixed appliance can effectively control the tooth movement in three-dimensional space, precisely adjust the teeth, and make the tooth rows into the ideal arch form. Combination of these two kinds of appliances can obtain the complementary efficiency advantages of the appliances, and shorten the treatment course. Expansion can cause mandibular clockwise rotation and mandibular plane angle increasing, while with the application of upper and lower fixed appliance, the occluding relation between maxil a and mandible can be adjusted more precisely in order to make the mandibular first molar move to the maxil ary molar buccal, establish good occlusal relationship, so that decrease the mandibular plane angle. So the rapid maxil ary expansion combined with fixed orthodontic can be used appropriately in the treatment of the patients with high angle intraversion.

ABSTRACT

Objective To study the asymmetry of the soft tissue thickness of upper eyelids in Shanghai female undergraduates undergoing double eyelid surgery. Methods Data were collected in 565 female students from Shanghai universities. Before surgery, oblique sagittal view images of the upper eyelids by 3.0T MRI were obtained and the following parameters were measured: soft tissue thickness at upper tarsal plate margin and upper central tarsal plate,sub-eyebrow and intra-orbital septum fat pad areas, and whole upper eyelid soft tissue areas. The resected orbicularis and intra-orbital septum fat were weighed by a highly accurated electronic balance. The eyelid asymmetry index (EAI) was calculated. Results The MRI (U/U2) central tarsal plate soft tissue thickness were:4.46±0.90 in the right side and 3.78±1.01 in the left; the intra orbital septum fat areas were:172.33±49.29 in the right and 136.34±37.42 in the left; the whole tissue areas were: 697.13±146.99 in the right and 500.66±158.87 in the left (P<0.01 for all). The weight of the resected orbicularis oculi muscle and intra orbital septum fat pad were (0.18±0.05) g and (0.17±0.06) g for the right side, and (0.15±0.04) g and (0.06±0.05) g for the left side (P<0.01 for all),respec tively. The orbicularis EAI was 0.17±0.06,and the intra orbital septum fat EAI was 0.41 ±0.08.Conclusions The asymmetric phenomenon of the upper eyelids' soft tissue thickness is commonly found in the Shanghai female undergraduates, and the main manifestation is that orbicularis and intra orbital septum fat in right side are thicker than that in the left.
